Bremelanotide is a synthetic cyclic heptapeptide developed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and appetite signalling. Its structure contains seven amino acid residues joined by a lactam bridge that closes the ring between two side chains. The molecular formula is C50H68N14O10 and the nominal molecular mass is near 1025 daltons. Much of the early laboratory literature refers to the same molecule by the development code PT-141.
The compound emerged from a research programme examining melanocortin analogues for effects on skin pigmentation. During early human studies, participants reported spontaneous erections as an unexpected side effect, which redirected development toward sexual function rather than tanning. An intranasal formulation was investigated in clinical trials but did not reach market approval. A subcutaneous injectable version later completed the regulatory process, and the nasal route does not appear in approved labelling.
The lactam ring that closes the peptide backbone improves resistance to exopeptidase attack, but the molecule stays susceptible to hydrolysis and oxidation once dissolved. Degradation accelerates with temperature, extreme pH, and light exposure, and repeated freeze-thaw cycles promote aggregation and material loss. Lyophilized powder held desiccated at or below minus twenty degrees Celsius is the common way to keep reference material. Reconstituted solutions are generally kept cold and used within a short window because their stability is far lower than that of the dry solid.
Handling procedures emphasize minimizing moisture uptake, since the dried solid is hygroscopic and can draw water when warmed to room temperature before a vial is opened. Laboratories record lot number, purity, counter-ion content, and residual solvent data supplied by the producer. Impurity profiles are compared run to run, and any shift in retention time or peak shape prompts re-verification against a reference standard. Certificates of analysis are treated as claims to be checked rather than accepted at face value.

Published studies differ in design, population and endpoint definition, so results are not always directly comparable across reports. Some trials used patient-reported measures of desire and distress, while others tracked physiological or behavioural outcomes. Questions that remain open include the durability of effects beyond short follow-up periods, the frequency of transient blood pressure elevation observed after administration, and whether a subtype-selective analogue could separate central effects from pigmentation-related activity. These points are usually framed as unresolved rather than settled in review literature.
Routine characterisation of bremelanotide relies on reversed-phase high-performance liquid chromatography with ultraviolet detection near 214 nm, where the peptide backbone absorbs. Mass spectrometry, typically in tandem mode with electrospray ionisation, confirms identity and supports quantification in biological matrices. Additional checks include amino acid analysis, peptide mapping after enzymatic digestion, and confirmation of the lactam bridge, since incomplete cyclisation produces a mass-shifted by-product. Purity values above 95 percent are common in reference-grade material, though reports vary in how strictly related substances are resolved from the parent peak.
The lyophilised solid is relatively stable when kept dry, protected from light and held at reduced temperature, commonly minus 20 degrees Celsius or lower for long-term storage. In solution the peptide is more vulnerable: tryptophan oxidation, hydrolysis of the lactam bridge and aggregation all become relevant over time, and the rate depends on pH, buffer composition and concentration. Repeated freeze-thaw cycles are generally avoided because they promote aggregation. Aqueous working solutions are typically prepared fresh or split into single-use aliquots to limit degradation before analysis.
Outside the approved product, bremelanotide circulates as a research chemical sold by peptide vendors, often labelled PT-141. Such material is not manufactured under pharmaceutical quality standards, and independent testing has repeatedly found content that differs from the label. Analytical certificates supplied with a purchase are not strong evidence of purity because they are usually generated by the seller. Online discussion tends to blur the distinction between the approved drug and unregulated powder, which complicates interpretation of reported experiences.
PT-141 is the original development code for bremelanotide, a synthetic peptide first studied as a potential tanning and sexual-response agent in the 1990s. Researchers at a small American biotechnology firm designed it as a shortened analogue of melanotan II, which itself came from work on alpha-melanocyte-stimulating hormone. Early screening focused on pigmentation, but behavioural observations in animal models redirected attention toward sexual motivation. That shift made PT-141 one of the first melanocortin compounds investigated specifically for effects on desire rather than on skin colour.
== Biological origin == Isoprenoid GDGTs originate as archaeal membrane lipids, whose fatty acids are converted to glycerol via esterification (ether lipid). They were first recognized as being associated with extremophilic archaea, but research in recent decades has discovered the compounds in a wide range of mesophilic environments as well, including soils, lake sediment, and marine deposits. Archaeal phylogenetic classes Nitrososphaerota (formerly Thaumarchaeota), Thermoproteota (formerly Crenarchaeota), "Euryarchaeota", and "Korarchaeota" produce GDGTs. Branched GDGTs are most commonly detected in peats and soils and are most associated with terrestrial settings. To date, no direct evidence for an unequivocal source organism has been reported, but the structural similarity of acidobacterial lipid to brGDGT alkyl chains strongly suggests that acidobacteriota synthesize brGDGT. The stereochemistry strongly hints at a non-archaeal origin.
Trump characterized the action as an application of the Monroe Doctrine, which he styled as the "Donroe Doctrine", and added that "American dominance in the western hemisphere will never be questioned again." Trump further stated that US oil companies would "go in, spend billions of dollars, fix the badly broken infrastructure, the oil infrastructure, and start making money for the country", and that the US was prepared to launch a second, much larger attack. He also expressed openness to a US occupation of Venezuela, stating it would not cost the US anything because it would be reimbursed through revenue from Venezuela's oil reserves. On 7 January, Trump stated that US efforts to revitalize Venezuela could take "much longer" than a year, adding that the Venezuelan authorities were doing "everything that we feel is necessary". Regarding the reward the US government offered for information leading to Maduro's capture, Rubio added that nobody will be receiving it. Rubio said "I guess we save $50 million" to which Trump replied "Don't let anybody claim it. Nobody deserves it but us." On 4 January 2026, Marco Rubio retracted Trump's comments of the day before, saying the US would not govern Venezuela, but would continue to enforce the existing oil quarantine in order to pressure Venezuela for policy changes. Rubio also affirmed that the US did not officially recognize Rodríguez's presidency, despite Trump's avowal that he would work with Rodríguez.
==== McLean v. Arkansas ==== In 1981, Kenyon was recruited to be an expert witness for the creationist side in the McLean v. Arkansas case that tested the constitutionality of Arkansas' Equal Time Legislation that mandated equal time for "creation science" and "evolution science". Kenyon flew to Arkansas to be deposed and testify during the trial. However, apparently under the influence of creationist attorney Wendell Bird (who was displeased with the defense of the creationist position by the Arkansas attorney general Steve Clark), Kenyon left town just before he was to testify:
== Cosmetics == Thickening agents used in cosmetics or personal hygiene products include viscous liquids such as polyethylene glycol, synthetic polymers such as carbomer (a trade name for polyacrylic acid) and vegetable gums. Some thickening agents may also function as stabilizers when they are used to maintain the stability of an emulsion. Some emollients, such as petroleum jelly and various waxes may also function as thickening agents in an emulsion.

Patients with heart disease, and patients who smoked cigarettes, were less likely to survive. Another trial, the Stem Cell Transplant vs. Cyclophosphamide (SCOT) trial, is ongoing. Asengeprast is an experimental systemic scleroderma drug candidate. It is a small molecule inhibitor of the G-protein coupled receptor GPR68 with antifibrotic activity.
=== Molecular mimicry === Some infectious agents, like Campylobacter jejuni, bear antigens that resemble, but are not identical to, the body's self-molecules. This phenomenon, known as molecular mimicry, can lead to cross-reactivity, where the immune response to such infections inadvertently results in the production of antibodies that also react with self-antigens. An example of this is Guillain–Barré syndrome, in which antibodies generated in response to a C. jejuni infection also react with the gangliosides in the myelin sheath of peripheral nerve axons.
Some nuclear safety agencies make recommendations about the use of thorium mantles and have raised safety concerns regarding their manufacture and disposal; the radiation dose from one mantle is not a serious problem, but that from many mantles gathered together in factories or landfills is.
Diacetyldihydromorphine (also known as Paralaudin, dihydroheroin, acetylmorphinol) is a potent opiate derivative developed in Germany in 1928 which is rarely used in some countries for the treatment of severe pain such as that caused by terminal cancer, as another form of diacetylmorphine (also commonly known as Heroin). Diacetyldihydromorphine is fast-acting and longer-lasting than diamorphine, with a duration of action of around 4–7 hours. As an ester/analogue of dihydromorphine, diacetyldihydromorphine is presumably a Schedule I/Narcotic controlled substance in the United States but does not have its own ACSCN or annual production quota. It does appear in the German Betäubungsmittelgesetz and other European controlled-substances laws. Diacetyldihydromorphine is quickly metabolized by plasma esterase enzymes into dihydromorphine, in the same way that diamorphine is metabolized into morphine. Diacetyldihydromorphine is roughly equipotent to morphine, where as diamorphine (heroin) is 1.50–1.80 times the potency of morphine. It shares with other opioids the risk of overdose or (potentially life-threatening) respiratory depression. When strong narcotics are required, and morphine and diamorphine are not an option, it is more common to use better known drugs such as nicomorphine, hydromorphone, levorphanol, oxymorphone or fentanyl which doctors will be more familiar with, and which do not share the stigma associated with either heroin or morphine.
In 1939, Drew married Minnie Lenore Robbins, a professor of home economics at Spelman College in Atlanta, Georgia, whom he had met earlier during that year. They had three daughters and a son. His daughter Charlene Drew Jarvis served on Council of the District of Columbia from 1979 to 2000, was the president of Southeastern University from 1996 until 2009, and was a president of the District of Columbia Chamber of Commerce.

He presented the mechanism of the β-lactam development with proton sponge as the stoichiometric base, and also discussed the kinetic analysis of the catalyzed reaction of alkenes with α-imino esters. Lectka has studied the transition-metal catalyzed amide isomerization and peptide folding. He presented the first spectroscopic and crystallographic proof of copper(II)-sodium coordination in tertiary amides and discussed the role of side chain in substituted prolines as a binding site for copper. Lectka's research during his term at Johns Hopkins University also focused on enantioselective halogenation, cooperative asymmetric catalysis, the medicinal chemistry of fluorinated molecules, and studies on asymmetric catalysis on sequentially-linked columns leading to synthesis machines. He conducted research on the chemistry of [C-F-C] fluoronium ions and later reported first spectroscopic evidence for fluoronium ions in a solution. Lectka has also worked on metal-catalyzed aliphatic fluorination and site-selective aliphatic fluorination. Lectka has also established the use of fluorine as a through-space activating substituent for aromatic substitution.
Attempts are being made to restore many of the city's best-maintained examples of pre-Soviet architecture. These restored structures are distinguished by bright colors and particularly clean façades. There are also a few examples of Soviet avant-garde work, such as the house of the architect Konstantin Melnikov in the Arbat area. Many of these restorations were criticized for allegedly disrespecting historical authenticity. In addition, facadism is widely practiced. Later examples of Soviet architecture are usually distinguished by large size and semi-Modernist styles. An example is the Novy Arbat project—informally called the "false teeth of Moscow"—and known for widespread disruption of the project's historic area in central Moscow.
== Subcellular distribution of isoforms == In mouse and rat, three distinct GPX4 isoforms with different subcellular localization are produced through alternative splicing and transcription initiation; cytosolic GPX4, mitochondrial GPX4 (mGPX4), and nuclear GPX4 (nGPX4). Cytosolic GPX4 has been identified as the only GPX4 isoform being essential for embryonic development and cell survival. The GPX4 isoforms mGPX4 and nGPX4 have been implicated in spermatogenesis and male fertility. In humans, experimental evidence for alternative splicing exists; alternative transcription initiation and the cleavage sites of the mitochondrial and nuclear transit peptides need to be experimentally verified.
=== Oncological research === In January 2026, a perspective piece in The EMBO Journal proposed mechanistic parallels between cancer cell behavior and helminth (parasitic) infections, specifically regarding immune evasion, migration, and immune modulation. The piece suggested that because cancer cells and helminths share these biological strategies, anthelmintic drugs like ivermectin may be candidates for therapeutic repurposing. The proposed mechanism involves utilizing the drug's known antiparasitic action to disrupt oncogenic pathways and potentially trigger immunogenic responses in the host environment. While these theoretical frameworks and preclinical models identify ivermectin as a potential adjunct in cancer therapy, clinical efficacy in humans has not been established, and the research remains in the investigational phase.
The British had no answer to the V-2 rocket bombardment that began in September 1944. The missiles flew too high and too fast to be detected during their approach, leaving no time even for an air raid warning to be sounded. Their supersonic speed meant that the explosions occurred without warning before the sound of their approach reached the target. The government initially tried to pass them off as explosions in the underground gas mains. It was clear this was not the case, and eventually, examples of the V-2 falling in its final plunge were captured on film. In response, several CH stations were re-organized into the "Big Ben" system to report the V-2s during launch. No attempt was made to try to find the location of the launch; the radio-goniometer was simply too slow to use. Instead, each of the stations in the network, Bawdsey, Gt. Bromley, High St, Dunkirk and Swingate (Dover) were left set to their maximum range settings and in the altitude measuring mode. In this mode, the radar had several stacked lobes where they were sensitive to signals. As the missile ascended it would pass through these lobes in turn, causing a series of blips to fade in and out over time. The stations attempted to measure the ranges to the target as they flew through each of these lobes and forwarded that by telephone to a central plotting station. At the station, these range measurements were plotted as arcs on a chart, known as range cuts. The intersections of the arcs defined the approximate area of the launcher.

It is a synthetic cyclic peptide that acts as an agonist at melanocortin receptors. It is not a hormone replacement therapy and not a vasodilator in the usual clinical sense, although it does affect vascular tone. Chemically it belongs to the melanocortin analogue family.
PT-141 was the internal development code used during early research and it persists in older publications, conference abstracts, and informal listings. The code and the approved name refer to the same molecule. Its continued use reflects habit in the literature rather than any difference in substance.
No. The intranasal form was studied in trials but was never approved, and absorption through the nasal route varies considerably between individuals. Approved labelling covers a subcutaneous injection only. Any nasal product sold outside regulated channels falls outside verified manufacturing.
Chemical degradation such as hydrolysis and oxidation proceeds more slowly as temperature falls. Cold storage also limits microbial growth in reconstituted material. Repeated warming should be avoided because thermal cycling stresses the peptide.
