Bremelanotide is a synthetic cyclic heptapeptide developed under the research code PT-141. The code reflects its position in an internal compound series rather than a chemical classification, and the name bremelanotide was later adopted for regulatory filings. Structurally it belongs to the melanocortin peptide family and shares a core sequence motif with alpha-melanocyte-stimulating hormone. The compound is supplied as an acetate salt in aqueous solution for injection. In reference literature it is indexed under both the code and the generic name, a dual listing that can complicate database searches.
Early work on melanocortin analogs in the 1980s and 1990s produced peptides intended to influence pigmentation and appetite. One of these, melanotan II, was observed to affect sexual desire as an incidental finding in self-administration reports. Researchers then pursued analogs with altered receptor selectivity and improved handling characteristics, and PT-141 emerged from that program in the late 1990s. The development path moved from dermatology and metabolism toward a central nervous system application, a shift that shaped both trial designs and the eventual label.
Regulatory review of bremelanotide concluded in 2019 with approval in the United States for a defined indication in premenopausal women. The reviewed formulation is a single-use prefilled autoinjector given subcutaneously, and its label carries cardiovascular monitoring language tied to blood pressure changes recorded during trials. Availability outside the approving jurisdiction varies, and in several countries the compound remains unapproved or is handled as a prescription-only item. Compounded and research-grade material also circulates, and it differs from the reviewed product in purity, characterization, and chain of custody.
Bremelanotide is a cyclic heptapeptide that acts as an agonist at melanocortin receptors. It binds MC1R, MC3R, MC4R, and MC5R, with MC4R activation considered most relevant to sexual desire pathways in the central nervous system. The molecule is a synthetic analog of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and energy regulation. Early research explored its use in tanning before attention shifted toward sexual dysfunction applications. Receptor binding affinity varies across these subtypes.
Activation of MC4R in the hypothalamus is thought to influence dopaminergic signaling, which in turn affects arousal and desire. This mechanism differs from that of phosphodiesterase type 5 inhibitors, which act primarily on vascular smooth muscle in the genital region. Because the pathway is central rather than peripheral, effects are not strictly dependent on local blood flow. The precise downstream cascade linking receptor binding to behavioral outcomes remains an area of ongoing investigation.

Compared with the related compound melanotan II, PT-141 shows markedly weaker activation of receptors tied to pigmentation. This difference stems from subtle structural variations that alter affinity distribution across receptor subtypes. Investigators propose that such selectivity produces a different side effect profile in specific applications. However, downstream consequences of prolonged receptor activation remain uncertain in the literature. Published studies do not fully agree on the duration of signaling pathway activity and the mechanisms of desensitization.
From a pharmacokinetic standpoint, the peptide is usually delivered by injection because oral bioavailability is very low; proteases in the digestive tract degrade it rapidly. After subcutaneous administration, plasma concentrations reach a peak within roughly one hour. Its elimination half-life is relatively short, with most reports placing it in the range of a few hours. Nasal formulations have also been examined, though absorption varies widely between individuals. Metabolism proceeds mainly through peptidase cleavage, and the resulting products are excreted by the kidneys.
Lyophilised peptide is generally held below minus twenty degrees Celsius, protected from light and moisture, because hydrolysis and oxidation accumulate faster at ambient temperature. Once reconstituted, solutions are typically kept between two and eight degrees Celsius and used within a short window defined by the supplier. Repeated freeze-thaw cycles are avoided since they promote aggregation and loss of soluble material. Container material matters as well, because peptides adsorb to certain plastics and glass surfaces at low concentration. Stability figures supplied by a vendor apply only to the specific lot and buffer that were tested.
Identity and purity are usually established with reversed-phase high-performance liquid chromatography combined with mass spectrometry. A gradient of water and acetonitrile containing trifluoroacetic acid is a common mobile phase, and ultraviolet detection near 214 nanometres responds to the peptide backbone. Mass spectrometry confirms the expected molecular mass and helps reveal truncation or oxidation products. Purity is reported as a peak-area percentage, a figure that depends on the wavelength and gradient used, so values from different laboratories are not always directly comparable. Peptide mapping and amino acid analysis provide additional confirmation when required.
Regulatory status varies by jurisdiction, where approved prescription products, compounded preparations and research-grade material are treated as distinct categories with different documentation requirements. Suppliers of research material commonly issue a certificate of analysis listing purity, identity and sometimes endotoxin content. Independent verification by a third-party laboratory is often recommended because self-reported figures are difficult to check. Literature discussions usually state the source, purity and storage conditions of the material used, since these details affect reproducibility. Analysts note that a reported purity figure does not by itself describe biological activity.
Clinical programmes in this area have relied mainly on randomised, double-blind, placebo-controlled designs in premenopausal women. Primary endpoints usually combine a validated questionnaire covering desire domains with counts of satisfying sexual events and a separate measure of distress. Reported outcomes show statistically significant but modest average improvement over placebo, with wide individual variation. Adverse events such as nausea, flushing, and headache occur frequently and can limit tolerability. Whether short-term trial gains translate into lasting change for most users is an open question.
Evidence outside the studied population is sparse. Trials have concentrated on premenopausal women with a defined diagnosis, and data for postmenopausal women, men, and people taking interacting medications remain limited. Non-prescription use of the peptide for comparable goals is widespread but is not supported by published controlled data. Observed changes in blood pressure have drawn attention to cardiovascular monitoring during use. The literature generally frames the compound as a targeted receptor agonist rather than a general libido enhancer, and basic questions about mechanism and long-term safety are unresolved.
Bremelanotide functions as an agonist at several melanocortin receptor subtypes, with the strongest functional activity reported at the MC4 subtype. MC4 receptors sit in hypothalamic circuits that influence appetite, energy balance, and components of sexual behaviour. Rodents lacking functional MC4 receptors show altered mating behaviour, which supports a role for this pathway in desire. The precise sequence of events connecting receptor activation to reported human effects remains only partly characterised. Because the same receptor family governs pigmentation and inflammatory signalling, selectivity is a recurring theme in pharmacological discussion.

=== Galactosemia === Galactosemia results from an inability to process galactose, a simple sugar. This deficiency occurs when the gene for galactose-1-phosphate uridylyltransferase (GALT) has any number of mutations, leading to a deficiency in the amount of GALT produced. There are two forms of Galactosemia: classic and Duarte. Duarte galactosemia is generally less severe than classic galactosemia and is caused by a deficiency of galactokinase. Galactosemia renders infants unable to process the sugars in breast milk, which leads to vomiting and anorexia within days of birth. Most symptoms of the disease are caused by a buildup of galactose-1-phosphate in the body. Common symptoms include liver failure, sepsis, failure to grow, and mental impairment, among others. Buildup of a second toxic substance, galactitol, occurs in the lenses of the eyes, causing cataracts. Currently, the only available treatment is early diagnosis followed by adherence to a diet devoid of lactose, and prescription of antibiotics for infections that may develop.
Aspartic, glutamic, and metallo-proteases activate a water molecule, which performs a nucleophilic attack on the peptide bond to hydrolyze it. Serine, threonine, and cysteine proteases use a nucleophilic residue (usually in a catalytic triad). That residue performs a nucleophilic attack to covalently link the protease to the substrate protein, releasing the first half of the product. This covalent acyl-enzyme intermediate is then hydrolyzed by activated water to complete catalysis by releasing the second half of the product and regenerating the free enzyme

== Upholstery filling == Buckwheat hulls are used as filling for a variety of upholstered goods, including pillows. The hulls are durable and do not insulate or reflect heat as much as synthetic filling. They are sometimes marketed as an alternative natural filling to feathers for those allergic to feathers. However, medical studies to measure the health effects of pillows manufactured with unprocessed and uncleaned hulls concluded that such pillows contain higher levels of a potential allergen that may trigger asthma in susceptible individuals than do synthetic-filled pillows.
MLSE and NBA had applied to register the Raptor's logo Raptors, a basketball with three diagonal claw gashes, for various goods and services. The Trademark Trial and Appeal Board dismissed Monster's claim. In March 2016, Monster filed a lawsuit to revoke the company trademark of Thunder Beast LLC of Washington, D.C., a small root beer brewery, insisting the use of "beast" in the company name encroached on Monster's trademark slogan, "Unleash the Beast". The owner of Thunder Beast, Stephen Norberg, was fighting Monster's lawsuit as of 2019. In August 2018, Monsta Pizza of Wendover in Buckinghamshire, United Kingdom, fought a claim brought by Monster Energy in the Intellectual Property Office. The claim was that the pizza firm could be mistaken for Monster Energy, leaving customers confused. The pizza firm won a landmark ruling against Monster Energy, when the court decided there would be no case of misrepresentation. In fact, Monster Energy loses many of their lawsuits; however, as with other cases of "trademark bullying", the main goal of the lawsuit seems to be bankrupting the opposition at which they are quite successful. Also in 2018, Monster Energy brought a lawsuit against Vital Pharmaceuticals Inc. (VPX), then producer of Bang Energy, a competitor of Monster Energy. The lawsuit alleged false advertising as well as theft of trade secrets. The matter concluded with a jury verdict in favor of Monster Energy.
==== Lay Low ==== In January 2015, Doillon started recording and co-producing, alongside Timber Timbre frontman Taylor Kirk, her second album in Canada. She recorded in the Montreal mythical studio Hotel2Tango, over a period of a month, with fellow band members of Timber Timbre. She completed the recording between Studio la Seine in Paris and RAK Studios in London. Lay Low was mixed by Nick Launey and Adam Greenspan in Los Angeles in April. The album came out on 9 October, preceded by the first single "Where to Start", which was featured in Jean-Marc Vallée's' movie Demolition (starring Naomi Watts and Jake Gyllenhaal) soundtrack. Doillon was nominated for best rock album of the year at Les Victoires de la musique. The album received critical acclaim in France, Belgium, England, Canada and the US and was released through Verve in the US and decca in the UK. The front cover for the album is a selfie shot by Doillon in bed, and inner pictures were shot during the recording in Canada by French photographer Zelie Noreda. In 2016, Doillon started a world tour, performing in the US, Canada, Japan, Australia, Korea, Germany, Poland, England and France. She performed on Jools Holland, alongside Iggy Pop and Graham Nash. As a final touch to her world tour, she performed two sold out concerts in La Flèche D'or, named Kitchen Acoustic and Lou Doillon and Friends (performed with fellow musicians Woodkid and Keren Ann).
Some ant species are considered as pests, primarily those that occur in human habitations, where their presence is often problematic. For example, the presence of ants would be undesirable in sterile places such as hospitals or kitchens. Some species or genera commonly categorized as pests include the Argentine ant, immigrant pavement ant, yellow crazy ant, banded sugar ant, pharaoh ant, red wood ant, black carpenter ant, odorous house ant, red imported fire ant, and European fire ant. Some ants will raid stored food, some will seek water sources, others may damage indoor structures, some may damage agricultural crops directly or by aiding sucking pests. Some will sting or bite. The adaptive nature of ant colonies make it nearly impossible to eliminate entire colonies and most pest management practices aim to control local populations and tend to be temporary solutions. Ant populations are managed by a combination of approaches that make use of chemical, biological, and physical methods. Chemical methods include the use of insecticidal bait which is gathered by ants as food and brought back to the nest where the poison is inadvertently spread to other colony members through trophallaxis. Management is based on the species and techniques may vary according to the location and circumstance.

SRIs like citalopram and paroxetine, as well as the serotonin 5-HT2A receptor antagonist ketanserin, have been found to partially block the increases in heart rate and blood pressure with MDMA. It is notable in this regard that serotonergic psychedelics such as psilocybin, which act as serotonin 5-HT2A receptor agonists, likewise have sympathomimetic effects. The NRI reboxetine and the serotonin–norepinephrine reuptake inhibitor (SNRI) duloxetine block MDMA-induced increases in heart rate and blood pressure. Conversely, bupropion, a norepinephrine–dopamine reuptake inhibitor (NDRI) with only weak dopaminergic activity, reduced MDMA-induced heart rate and circulating norepinephrine increases but did not affect MDMA-induced blood pressure increases. On the other hand, the robust NDRI methylphenidate, which has sympathomimetic effects of its own, has been found to augment the cardiovascular effects and increases in circulating norepinephrine and epinephrine levels induced by MDMA. The non-selective beta blocker pindolol blocked MDMA-induced increases in heart rate but not blood pressure. The α2-adrenergic receptor agonist clonidine did not affect the cardiovascular effects of MDMA, though it reduced blood pressure. The α1-adrenergic receptor antagonists doxazosin and prazosin blocked or reduced MDMA-induced blood pressure increases but augmented MDMA-induced heart rate and cardiac output increases. The dual α1- and β-adrenergic receptor blocker carvedilol reduced MDMA-induced heart rate and blood pressure increases.
On 2 November 2010, Lions Gate Entertainment secured the North American distribution rights to Dredd. Headey joined the cast as drug-dealer Ma-Ma in January 2011. Judge Dredd creator John Wagner acted as a consultant on the film. PT-141 was the internal development code assigned to bremelanotide during preclinical research. Reference sources sometimes index the peptide under the code rather than the generic name. The two terms describe the same molecule.
They are related but distinct cyclic peptides. Melanotan II is an earlier and less selective melanocortin analog. Bremelanotide was developed later with a narrower receptor profile and a defined clinical application.
The reviewed product is given by subcutaneous injection using a single-use autoinjector. Administration occurs under prescription supervision. The device delivers one fixed amount per use rather than a measured or adjustable quantity.
PT-141 is the research code for bremelanotide, a cyclic peptide developed as a melanocortin receptor agonist. The code has appeared in literature and catalog listings since early development. Bremelanotide is the international nonproprietary name.
