After subcutaneous administration, plasma concentrations rise within roughly thirty minutes and the elimination half-life is short, on the order of two to three hours. Reported physiological responses include transient increases in blood pressure and nausea, which tended to diminish with repeated dosing in trial settings. Because the peptide clears quickly, effects are not expected to persist long after a dose. Absorption from non-injected routes is poorly characterised, and nasal delivery produced variable plasma levels in older work.
Clinical research typically uses randomised, double-blind, placebo-controlled designs. The most common primary endpoint is the desire domain score of the Female Sexual Function Index, sometimes paired with a distress measure. Secondary outcomes include arousal, satisfaction, and event-based counts of satisfying sexual episodes. Across trials, average improvements are modest and individual responses vary widely. Whether benefits persist beyond a few months, and whether they depend on baseline hormone status, remain open questions rather than settled findings.
Approved use is narrow and jurisdiction-specific. In the United States, the injectable product is authorised for premenopausal women with acquired, generalised hypoactive sexual desire disorder, a diagnosis that requires documented distress. It is not approved for men, for postmenopausal women, or for use alongside hormonal contraceptives under the approved labelling. Outside regulated markets, the same peptide is frequently sold as a research chemical, where identity, purity, and sterility are not independently verified.
Bremelanotide is a synthetic cyclic heptapeptide developed as an analogue of alpha-melanocyte-stimulating hormone, a naturally occurring peptide involved in pigmentation and appetite signalling. Its structure contains seven amino acid residues joined by a lactam bridge that closes the ring between two side chains. The molecular formula is C50H68N14O10 and the nominal molecular mass is near 1025 daltons. Much of the early laboratory literature refers to the same molecule by the development code PT-141.
The compound emerged from a research programme examining melanocortin analogues for effects on skin pigmentation. During early human studies, participants reported spontaneous erections as an unexpected side effect, which redirected development toward sexual function rather than tanning. An intranasal formulation was investigated in clinical trials but did not reach market approval. A subcutaneous injectable version later completed the regulatory process, and the nasal route does not appear in approved labelling.

Identity and purity are usually established with reversed-phase high-performance liquid chromatography combined with mass spectrometry. A gradient of water and acetonitrile containing trifluoroacetic acid is a common mobile phase, and ultraviolet detection near 214 nanometres responds to the peptide backbone. Mass spectrometry confirms the expected molecular mass and helps reveal truncation or oxidation products. Purity is reported as a peak-area percentage, a figure that depends on the wavelength and gradient used, so values from different laboratories are not always directly comparable. Peptide mapping and amino acid analysis provide additional confirmation when required.
Regulatory status varies by jurisdiction, where approved prescription products, compounded preparations and research-grade material are treated as distinct categories with different documentation requirements. Suppliers of research material commonly issue a certificate of analysis listing purity, identity and sometimes endotoxin content. Independent verification by a third-party laboratory is often recommended because self-reported figures are difficult to check. Literature discussions usually state the source, purity and storage conditions of the material used, since these details affect reproducibility. Analysts note that a reported purity figure does not by itself describe biological activity.
Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.
PT-141 is the research code for bremelanotide, a cyclic heptapeptide derived from alpha-melanocyte-stimulating hormone. The molecule belongs to the melanocortin receptor agonist family and shows highest affinity for the MC4 receptor subtype, with weaker activity at MC1, MC3 and MC5. Its structure retains the core His-Phe-Arg-Trp sequence that defines melanocortin recognition, while cyclization and terminal modifications improve metabolic stability relative to the parent hormone. Early work classified the compound as a centrally acting agent rather than a peripherally acting vasodilator, which shaped subsequent development priorities.
==== Annexation of Subcarpathian Ruthenia by the Soviet Union ==== In October 1944, Subcarpathian Ruthenia was taken by the Soviets. A Czechoslovak delegation under František Němec was dispatched to the area. The delegation was to mobilize the liberated local population to form a Czechoslovak army and to prepare for elections in cooperation with recently established national committees. Loyalty to a Czechoslovak state was tenuous in Carpathian Ruthenia. Beneš's proclamation of April 1944 excluded former collaborationist Hungarians, Germans and the Rusynophile Ruthenian followers of Andrej Bródy and the Fencik Party (who had collaborated with the Hungarians) from political participation. This amounted to approximately 1⁄3 of the population. Another 1⁄3 was communist, leaving 1⁄3 of the population presumably sympathetic to the Czechoslovak Republic. Upon arrival in Subcarpathian Ruthenia, the Czechoslovak delegation set up headquarters in Khust, and on 30 October issued a mobilization proclamation. Soviet military forces prevented both the printing and the posting of the Czechoslovak proclamation and proceeded instead to organize the local population. Protests from Beneš's government were ignored. Soviet activities led much of the local population to believe that Soviet annexation was imminent. The Czechoslovak delegation was also prevented from establishing a cooperative relationship with the local national committees promoted by the Soviets.
=== Food industry === The calcium sulfate hydrates are used as a coagulant in products such as tofu. For the FDA, it is permitted in cheese and related cheese products; cereal flours, bakery products, frozen desserts, artificial sweeteners for jelly & preserves, condiment vegetables, and condiment tomatoes, and some candies. It is known in the E number series as E516, and the UN's FAO knows it as a firming agent, a flour treatment agent, a sequestrant, and a leavening agent.
=== European Union === In August 2021, Foreign Minister Béjar announced Peru's withdrawal, without officially carrying it out, from the Lima Group and support for the International Contact Group of the European Union for a negotiated solution between the regime of Nicolás Maduro and the opposition regarding the Venezuelan crisis. In addition, Béjar announced his intention, as foreign minister, to reconstitute the UNASUR and strengthen the CELAC, although this was not officially carried out.In September 2021, the Peruvian government signed the "Development Objectives Grant Agreement" with the United States Agency for International Development (USAID) to receive 321 million dollars through 2026. The agreement was signed between Foreign Minister Maúrtua and Jene Thomas, USAID mission director, in the presence of U.S. ambassador Lisa Kenna, who before joining the diplomatic service had been an agent of the CIA. During that same month, the Peruvian government signed the United Nations Sustainable Development Cooperation Framework in Peru 2022-2026 for 180 million dollars, with the framework's preparation led by the resident coordinator of the United Nations System (UNS) in the country to "contribute to the efforts of the Government of Peru in fulfilling the 2030 Agenda and the Sustainable Development Goals, as well as the national development priorities with which this roadmap aligns", also establishing "the general guidelines for developing joint work plans between the Government of Peru and twenty-two AFPs [UN Agencies, Funds and Programmes]" over the following five years.
Furthermore, a deficiency in GIP receptors has been associated in mice with a dramatic decrease in bone quality and a subsequent increase in fracture risk. However, the results of the two groups' studies were non-concordant.
Codeine is an opiate and prodrug of morphine mainly used to treat pain, coughing, and diarrhea. It is commonly used as a recreational drug. It is found naturally in the sap of the opium poppy, Papaver somniferum. It is typically used to treat mild to moderate degrees of pain. Greater benefit may occur when combined with paracetamol (acetaminophen) as codeine/paracetamol or a nonsteroidal anti-inflammatory drug (NSAID) such as aspirin or ibuprofen. Evidence does not support its use for acute cough suppression in children. In Europe, it is not recommended as a cough medicine for those under 12 years of age. It is generally taken orally. It usually starts working after half an hour, with maximum effect at two hours. Its effects last for about four to six hours. Codeine exhibits abuse potential similar to other opioid medications, including a risk of addiction and overdose. Common side effects include nausea, vomiting, constipation, itchiness, lightheadedness, and drowsiness. Serious side effects may include breathing difficulties and addiction. Whether it can be used safely during pregnancy is unclear. Care should be used during breastfeeding, as it may result in opiate toxicity in the baby. Its use as of 2016 is not recommended for children. Codeine works by being broken down by the liver into morphine; how quickly this occurs depends on a person's genetics. Codeine was discovered in 1832 by Pierre Jean Robiquet. In 2013, about 361,000 kg (795,000 lb) of codeine were produced while 249,000 kg (549,000 lb) were used, which made it the most commonly taken opiate.

== Side effects == Side effects of fentanyl analogs are similar to those of fentanyl itself, which include itching, nausea and potentially serious respiratory depression, which can be life-threatening. Fentanyl analogs have killed hundreds of people throughout Europe and the former Soviet republics since the most recent resurgence in use began in Estonia in the early 2000s, and novel derivatives continue to appear. A new wave of fentanyl analogues and associated deaths began in around 2014 in the US, and have continued to grow in prevalence; especially since 2016 these drugs have been responsible for hundreds of overdose deaths every week.
The new name was proposed after 70% of a panel of experts expressed support for this name. This new name was adopted in 2023. Formerly, abnormal accumulation of fat in the liver in the absence of secondary causes of fatty liver, such as significant alcohol use, viral hepatitis, or medications that can induce fatty liver, was the definition of NAFLD. However, the term MASLD accepts that there may be other conditions present, but focuses on the metabolic abnormalities contributing to the disorder.
== Tissue expression == Alpha-synuclein is a synuclein protein primarily found in neural tissue, making up as much as one percent of all proteins in the cytosol of brain cells. It is expressed highly in neurons within the frontal cortex, hippocampus, striatum, and olfactory bulb, but can also be found in the non-neuronal glial cells. It has been established that alpha-synuclein is extensively localized in the nucleus of mammalian brain neurons, suggesting a role of alpha-synuclein in the nucleus. Synuclein is however found predominantly in the presynaptic termini, in both free or membrane-bound forms, with roughly 15% of synuclein being membrane-bound at any moment in neurons. It has also been shown that alpha-synuclein is localized in neuronal mitochondria. Alpha-synuclein is highly expressed in the mitochondria in olfactory bulb, hippocampus, striatum and thalamus, where the cytosolic alpha-synuclein is also rich. However, the cerebral cortex and cerebellum are two exceptions, which contain rich cytosolic alpha-synuclein but very low levels of mitochondrial alpha-synuclein. It has been shown that alpha-synuclein is localized in the inner membrane of mitochondria, and that the inhibitory effect of alpha-synuclein on complex I activity of the mitochondrial respiratory chain is dose-dependent. Thus, it is suggested that alpha-synuclein in mitochondria is differentially expressed in different brain regions and the background levels of mitochondrial alpha-synuclein may be a potential factor affecting mitochondrial function and predisposing some neurons to degeneration.

== Use and toxicity == The European species did not enter into the herbalists' pharmacopeia. In the American Old West, the Western white clematis, Clematis ligusticifolia, was called pepper vine by early travelers and pioneers, who took a tip from Spanish colonials and used seeds and the acrid leaves of yerba de chivato as a pepper substitute. The entire genus contains essential oils and compounds which are extremely irritating to the skin and mucous membranes. Unlike black pepper or Capsicum, however, the compounds in clematis cause internal bleeding of the digestive tract if ingested in large amounts. C. ligusticifolia is essentially toxic. When pruning them, it is a good idea to wear gloves. Despite its toxicity, Native Americans used very small amounts of clematis as an effective treatment for migraine headaches and nervous disorders. It was also used as an effective treatment of skin infections. Clematis is also a constituent of Bach's Rescue Remedy. Leaf extracts from two Ethiopian species (Clematis longicauda steud ex A. Rich. and Clematis burgensis Engl.) are used locally to treat ear disorders and eczema. Phytochemical screening of the extracts from both of these species showed antibacterial and antifungal activity. The extracts of these plants also possess wound healing and anti-inflammatory activities which could also be attributed to the phytoconstituents. Clematis has been listed as one of the 38 plants used to prepare Bach flower remedies, a kind of alternative medicine promoted for its effect on health.
) the proton lifetimes results to be far too short. To forbid these operators, a new symmetry has to be imposed: the R-parity. This symmetry also stabilizes the lightest supersymmetric particle as a dark matter candidate.
The Mesopotamians seem to have had little interest in the natural world as such, preferring to study how the gods had ordered the universe. Animal physiology was studied for divination, including especially the anatomy of the liver, seen as an important organ in haruspicy. Animal behavior too was studied for divinatory purposes. Most information about the training and domestication of animals was probably transmitted orally, but one text dealing with the training of horses has survived. The ancient Mesopotamians had no distinction between "rational science" and magic. When a person became ill, doctors prescribed both magical formulas to be recited and medicinal treatments. The earliest medical prescriptions appear in Sumerian during the Third Dynasty of Ur (c. 2112 – c. 2004 BCE). The most extensive Babylonian medical text, however, is the Diagnostic Handbook written by the ummânū, or chief scholar, Esagil-kin-apli of Borsippa, during the reign of the Babylonian king Adad-apla-iddina (1069 – 1046 BCE). In East Semitic cultures, the main medicinal authority was an exorcist-healer known as an āšipu. The profession was passed down from father to son and was held in high regard. Of less frequent recourse was the asu, a healer who treated physical symptoms using remedies composed of herbs, animal products, and minerals, as well as potions, enemas, and ointments or poultices. These physicians, who could be either male or female, also dressed wounds, set limbs, and performed simple surgeries.
Although the number of local news media outlets had increased in the 2000s, the Ba'ath Party continued to maintain its tight control and censorship of the press. Journalists and bloggers have been arrested and tried. In 2009, the committee to Protect Journalists named Syria number three in a list of the ten worst countries in which to be a blogger, given the arrests, harassment, and restrictions which online writers in Syria faced. Internet censorship in Ba'athist Syria was extensive. The Ba'athist government banned websites over political reasons and arrests people accessing them. Internet cafes are required to record all the comments users post on chat forums. Websites such as Wikipedia Arabic, YouTube and Facebook were blocked from 2008 to 2011. Filtering and blocking was found to be pervasive in the political and Internet tools areas, and selective in the social and conflict/security areas by the OpenNet Initiative in August 2009. Syria has been on Reporters Without Borders' Enemy of the Internet list since 2006 when the list was established. In addition to filtering a wide range of Web content, the Ba'athist Syrian government monitored Internet use very closely and detained citizens "for expressing their opinions or reporting information online." Vague and broadly worded laws invite government abuse and have prompted Internet users to engage in self-censorship to avoid the state's ambiguous grounds for arrest. The Syrian Centre for Media and Free Expression was closed by the Ba'athist government in September 2009.

Trial results generally show a small to moderate average improvement in desire scores relative to placebo. The distribution of responses is wide, and some participants show little measurable change. Group averages should not be read as a prediction for any single person.
Nausea, flushing, headache, and transient blood pressure elevation are the most frequently reported events. These typically appear shortly after dosing and are usually described as mild to moderate in severity. Safety data covering long periods of continuous use remain limited.
A short half-life means the compound clears from circulation within hours, so any effect is tied closely to dosing time. Accumulation between doses is therefore unlikely under the studied schedule. The practical consequence is that timing of administration shapes what observers record.
It is a synthetic cyclic peptide that acts as an agonist at melanocortin receptors. It is not a hormone replacement therapy and not a vasodilator in the usual clinical sense, although it does affect vascular tone. Chemically it belongs to the melanocortin analogue family.
