This is a working overview of melanocortin receptor, written for readers who want more than a one-paragraph summary but less than a textbook.
Reviewed 2025-09-17. Anything still debated is marked as such rather than presented as settled.
Bremelanotide functions as an agonist at several melanocortin receptor subtypes, with the strongest functional activity reported at the MC4 subtype. MC4 receptors sit in hypothalamic circuits that influence appetite, energy balance, and components of sexual behaviour. Rodents lacking functional MC4 receptors show altered mating behaviour, which supports a role for this pathway in desire. The precise sequence of events connecting receptor activation to reported human effects remains only partly characterised. Because the same receptor family governs pigmentation and inflammatory signalling, selectivity is a recurring theme in pharmacological discussion.
Clinical programmes in this area have relied mainly on randomised, double-blind, placebo-controlled designs in premenopausal women. Primary endpoints usually combine a validated questionnaire covering desire domains with counts of satisfying sexual events and a separate measure of distress. Reported outcomes show statistically significant but modest average improvement over placebo, with wide individual variation. Adverse events such as nausea, flushing, and headache occur frequently and can limit tolerability. Whether short-term trial gains translate into lasting change for most users is an open question.
Evidence outside the studied population is sparse. Trials have concentrated on premenopausal women with a defined diagnosis, and data for postmenopausal women, men, and people taking interacting medications remain limited. Non-prescription use of the peptide for comparable goals is widespread but is not supported by published controlled data. Observed changes in blood pressure have drawn attention to cardiovascular monitoring during use. The literature generally frames the compound as a targeted receptor agonist rather than a general libido enhancer, and basic questions about mechanism and long-term safety are unresolved.
Melanocortin receptors form a family of five G-protein-coupled receptors designated MC1 through MC5. Bremelanotide binds most strongly at MC4R and MC1R, with weaker activity reported at MC3R and MC5R. MC4R is expressed in hypothalamic nuclei that coordinate energy balance and aspects of sexual behaviour. The prevailing interpretation is that central MC4R activation, rather than peripheral vascular effects alone, drives the reported changes in desire. This account remains partly inferential, since direct receptor-level measurement in living humans is not practical.
After subcutaneous administration, plasma concentrations rise within roughly thirty minutes and the elimination half-life is short, on the order of two to three hours. Reported physiological responses include transient increases in blood pressure and nausea, which tended to diminish with repeated dosing in trial settings. Because the peptide clears quickly, effects are not expected to persist long after a dose. Absorption from non-injected routes is poorly characterised, and nasal delivery produced variable plasma levels in older work.
Clinical research typically uses randomised, double-blind, placebo-controlled designs. The most common primary endpoint is the desire domain score of the Female Sexual Function Index, sometimes paired with a distress measure. Secondary outcomes include arousal, satisfaction, and event-based counts of satisfying sexual episodes. Across trials, average improvements are modest and individual responses vary widely. Whether benefits persist beyond a few months, and whether they depend on baseline hormone status, remain open questions rather than settled findings.
| Property | Value | Notes |
|---|---|---|
| Primary receptor target | MC4 melanocortin receptor | Other melanocortin subtypes are also activated |
| Typical route in approved use | Subcutaneous injection | Single-use format in the marketed product |
| Elimination half-life | Roughly two to three hours | Short exposure supports as-needed use |
| Common adverse events | Nausea, flushing, headache | Generally transient but can be dose limiting |
| Main studied group | Premenopausal women with low desire | Controlled data outside this group are limited |
After subcutaneous dosing, peak plasma concentrations appear within roughly one hour, and elimination is fast, with a half-life on the order of a few hours. Degradation is mainly proteolytic, and at least one circulating fragment retains receptor activity, so parent-drug levels alone do not describe total exposure. Clearance does not depend heavily on hepatic cytochrome enzymes, which lowers the likelihood of common metabolic interaction routes. Data in renal or hepatic impairment are limited. Repeated dosing does not appear to produce marked accumulation given the short half-life.
Reported pharmacodynamic effects include transient rises in blood pressure and heart rate, flushing, nausea and headache, appearing soon after dosing and resolving within hours. These responses were dose-related in early studies and shaped the label's cardiovascular cautions and blood pressure monitoring advice. Gastrointestinal upset is the most frequent reason cited for discontinuation in trials. Whether the vascular signal attenuates with repeated use is not settled. Central effects on desire are described as emerging over weeks rather than immediately, which points to a cumulative rather than acute process.
From a pharmacokinetic standpoint, the peptide is usually delivered by injection because oral bioavailability is very low; proteases in the digestive tract degrade it rapidly. After subcutaneous administration, plasma concentrations reach a peak within roughly one hour. Its elimination half-life is relatively short, with most reports placing it in the range of a few hours. Nasal formulations have also been examined, though absorption varies widely between individuals. Metabolism proceeds mainly through peptidase cleavage, and the resulting products are excreted by the kidneys.
PT-141 initiates cellular signaling by binding to specific subtypes within the melanocortin receptor family. These receptors belong to the G protein-coupled receptor superfamily, and activation raises intracellular cyclic adenosine monophosphate levels. This cascade ultimately influences neuronal circuits in the central nervous system that are associated with sexual desire and arousal. Research indicates the compound's action concentrates in hypothalamic regions rather than peripheral tissues, which helps explain some observed pharmacological features. The selectivity of receptor binding underlies its functional differences.
Compared with the related compound melanotan II, PT-141 shows markedly weaker activation of receptors tied to pigmentation. This difference stems from subtle structural variations that alter affinity distribution across receptor subtypes. Investigators propose that such selectivity produces a different side effect profile in specific applications. However, downstream consequences of prolonged receptor activation remain uncertain in the literature. Published studies do not fully agree on the duration of signaling pathway activity and the mechanisms of desensitization.
The war has severely weakened Yemen’s health system. More than half of the country’s health facilities have been reported as non-functional, a situation attributed to airstrikes, damage to buildings, departures or non-payment of staff, and long-term shortages of medicines and equipment.Facilities that continue to operate frequently do so under difficult conditions, facing unreliable electricity, scarce fuel for generators and obstacles in maintaining hygiene standards or cold-chain storage. Researchers link these operation difficulties to port restrictions, disrupted supply chains and the broader economic crisis. This institutional collapse has coincided with a series of major epidemics. Yemen has experienced what many researchers describe as the largest recorded cholera outbreak in modern times, with over one million suspected cases and earlier World Health Organization estimates of more than 600,000 cases with over 2,000 deaths. The spread of cholera has been associated with the destruction of water treatment plants, malfunctioning sewage systems and fuel shortages that hindered water pumping and waste disposal. Interruptions in vaccination campaigns and the wider deterioration of primary health care also facilitated outbreaks of diphtheria and measles, while when the COVID-19 pandemic reached Yemen, observers reported delayed vaccine delivery, limited testing capacity and additional economic pressure due to rising prices and economic stagnation.
He returned in round 7, but had a subsequent dip in form. However, Essendon coach, Matthew Knights, persisted with Jetta and was paid back by his outstanding performance against Fremantle in round 14. The 2009 season was another important year in Jetta's development. A few quiet performances in the pre-season led to his omission from the side for the opening two rounds of the season. However, strong form for the Bendigo Bombers led to a recall for Essendon's vital round 3 encounter with Carlton. On 7 September 2014, Jetta parted ways with Essendon after eight seasons with the club. Jetta, along with 33 other Essendon players, was found guilty of using a banned performance-enhancing substance, thymosin beta-4, as part of Essendon's sports supplements program during the 2012 season. He and his teammates were initially found not guilty in March 2015 by the AFL Anti-Doping Tribunal, but a guilty verdict was returned in January 2016 after an appeal by the World Anti-Doping Agency. He was suspended for two years which, with backdating, ended in February 2017; as a result, he served approximately seventeen months of his suspension and missed the entire 2016 WAFL season and parts of the 2015/16 and 2016/17 NTFL seasons. He later played with Peel Thunder in the WAFL and played for Pinjarra in the WA Amateur Football League in 2018.
The primary purpose of a tablet counter (also known as a pill counter or drug counter) is to accurately count prescription medications in tablet or capsule form to aid the requirement for patient medication safety, to increase efficiency and reduce costs for the typical pharmacy. Newer versions of this counting device include advanced software to continue to improve safety for the patient who is receiving the prescription, ensuring that the pharmacy staff dispense the right medication at correct dosage strength for the right patient. (see also medication safety). Today's pharmacy industry recognizes the need for heightened vigilance against medication errors across the entire spectrum. A wealth of research has been conducted regarding the prevalence of medication errors and the ability of technology to decrease or eliminate such errors. (See the March 2003 landmark study by Auburn University's Center for Pharmacy Operations and Designs). Prescription dispensing safety and accuracy in the pharmacy are an essential part of ensuring the right patient gets the right medication at the right dosage. A trend in pharmacy is to place a greater reliance on technology and pharmacy automation to minimize the chance of human error and speed up the process of dispensing. Pharmacy management generally sees technology as a solution to industry challenges like staffing shortages, prescription volume increases, long and hectic work hours and complicated insurance reimbursement procedures.

=== Freeze drying === In planning for the long-duration Apollo missions, NASA conducted extensive research into space food. One of the techniques developed in 1938 by Nestlé was freeze drying. The foods are cooked, quickly frozen, and then slowly heated in a vacuum chamber to remove the ice crystals formed by the freezing process. The final product retains 98% of its nutrition and weighs much less than before drying. The ratio of weight before and after drying depends strongly on the particular food item but a typical freeze-dried weight is 20% of the original weight. In the United States, Action Products later commercialized this technique for other foods, concentrating on snack food resulting in products like Space ice cream. Today, one of the benefits of this advancement in food preservation includes simple, nutritious meals available to disabled and otherwise homebound senior adults unable to take advantage of existing meal programs.
Between 1634 and 1637, the enthusiasm for the new flowers in the Netherlands triggered a speculative frenzy now known as the tulip mania, the first recorded speculative bubble in history, that eventually led to the collapse of the market three years later. Tulip bulbs had become so expensive that they were treated as a form of currency, or rather, as futures, forcing the Dutch government to introduce trading restrictions on the bulbs. The 'Semper Augustus' was the most expensive tulip during the 17th-century tulip mania. At the height of 'Tulip mania' this tulip was even sold for 10.000 guilders (equivalent to approx. €130.000 in 2024). After seeing the tulip in the garden of Dr. Adriaan Pauw, a director of the Dutch East India Company, Nicolas van Wassenaer wrote in 1624 that "The colour is white, with carmine on a blue base, and with an unbroken flame right to the top". The 'Semper Augustus' was actually not a tulip variety, but rather a tulip affected by the tulip mosaic virus. With limited specimens in existence at the time and most owned by Pauw, his refusal to sell any flowers, despite wildly escalating offers, is believed by some to have sparked the mania. Around this time, the ceramic tulipiere was devised for the display of cut flowers stem by stem. Vases and bouquets, usually including tulips, often appeared in Dutch still-life painting. To this day, tulips are associated with the Netherlands, and the cultivated forms of the tulip are often called "Dutch tulips". The Netherlands has the world's largest permanent display of tulips at the Keukenhof.
During conflict, individuals receiving nasally administered oxytocin demonstrate more frequent defense-motivated responses toward in-group members than out-group members. Further, oxytocin was correlated with participants' desire to protect vulnerable in-group members, despite that individual's attachment to the conflict. Similarly, it has been demonstrated that when oxytocin is administered, individuals alter their subjective preferences to align with in-group ideals over out-group ideals. These studies demonstrate that oxytocin is associated with intergroup dynamics. Further, oxytocin influences the responses of individuals in a particular group to those of another group. The in-group bias is evident in smaller groups; however, it can also extend to groups as large as one's entire country, leading to a strong national zeal. A study done in the Netherlands showed that oxytocin increased the in-group favoritism of their nation while decreasing acceptance of members of other ethnicities and foreigners. People also show more affection for their country's flag while remaining indifferent to other cultural objects when exposed to oxytocin. It has thus been hypothesized that this hormone may be a factor in xenophobic tendencies secondary to this effect. Thus, oxytocin appears to affect individuals at an international level where the in-group becomes a specific "home" country, and the out-group grows to include all other countries.
It has been proposed to implant magnets within the body to overcome this limitation. Magnet location placement can be upstream or downstream of the location of the disease for maximum accumulation. Another question arises from the subject of cellular uptake. While the use of a magnetic field may guide particles to therapeutic sites, it is not an indicator of cellular uptake of particles. This raises questions regarding the effect, if any, of the therapeutic element on the effector area. Generally, nanoparticles efficiently cross cell barriers; however, this can change in the presence of other processes. Promise has been shown with the use of PEG coating. Hydrophilic coatings have shown enhanced cellular uptake at tumor cells with the use of a magnetic field. Another concern arises regarding the biotoxicity of magnetic nanoparticles. It is difficult to say for certain that all magnetic nanoparticles are toxic due the large variety of magnetic particles that can be used. The nanoparticles size, biodegradability, composition, and dosage are a few of the properties impacting this concern. However, it has been shown that magnetic nanoparticles that are either inhaled to enter the lungs or are swallowed and enter the gastrointestinal tract have unsatisfactory impacts on the body. PEG, linear neutral polyether coatings have a tendency to lose their targeting capabilities in response to their "immune stealthing" function.

Animosity between the Allies of World War II predated the end of the war. The death of Franklin D. Roosevelt in April 1945 and the changeover to Harry Truman's presidential administration led to changes in US-Soviet relations as Truman and his staff began to take a harder diplomatic line. US and Soviet diplomats disputed conflicting interpretations of agreements made between Stalin and Roosevelt at the Yalta Conference. Truman's administration was divided on the question of the Polish government's composition, with Truman and several cabinet members advocating for stronger demands that broader democratic elements be included in its ranks. A 1944 accord between Churchill and Stalin further complicated matters in Eastern Europe; the two had agreed to non-interference in one another's spheres of influence, in Greece and in Romania and Bulgaria respectively. The creation of the United Nations in the spring of 1945 led to further disagreements over how the postwar international order should be governed. Because of the alignment of many Latin American nations with the United States, the Soviets feared that a pro-American majority would be used against them. They opposed the admission of Argentina into the UN, and called for a more expansive Security Council veto. Stalin ultimately accommodated the American positions on the UN veto and on the Polish government, allowing Oskar Lange and Stanisław Mikołajczyk to join it.
=== Books === In 2003, Pain Killer: An Empire of Deceit and the Origin of America's Opioid Epidemic by the journalist Barry Meier was published. In 2023, Netflix adapted the book into the drama miniseries Painkiller. In 2015, Dreamland: The True Tale of America's Opiate Epidemic by Sam Quinones was released, which connected the growth of the U.S. opioid epidemic to the production and distribution of black-tar heroin from Mexico. In 2018, Dopesick: Dealers, Doctors, and the Drug Company that Addicted America by Beth Macy was released. The book covered the origin and evolution of the opioid epidemic in the United States beginning primarily with the 1996 release of the drug OxyContin, examining on its effects on small town America and the Appalachian region in particular. It received the Los Angeles Times Book Prize for Science and Technology. In 2021, Hulu adapted the book into a drama miniseries, also titled Dopesick, starring Michael Keaton. In 2021, Empire of Pain: The Secret History of the Sackler Dynasty by Patrick Radden Keefe was published, examining the history of the Sackler family, including the founding of Purdue Pharma, its role in the marketing of pharmaceuticals, and the family's central role in the opioid epidemic. The book followed Keefe's 2017 article on the Sackler family in The New Yorker, titled "The Family That Built an Empire of Pain". The book received multiple awards and was longlisted for the 2022 Andrew Carnegie Medal for Excellence in Nonfiction. In 2023, Netflix adapted Keefe's article and Meier's book into the drama miniseries Painkiller.
== Vegetable biomass-based textiles == Polysaccharide-based polymers are made of fruit and vegetable origin, and the most promising biopolymer-yielding fibers include cellulose, starch, chitin/chitosan, pectin, alginate, and carrageenan. Soy, whey, and zein could also be obtained from vegetable sources. An example is Pinatex, a pineapple-based leather that is manufactured from the cellulose fibers of pineapple leaves. It is an environmentally friendly alternative to traditional leather due to its sustainability and adaptability. Additionally its lightweight, resilient and biodegradable nature has made it very popular for new and innovative fashion.

== Use == The juice is drinkable, yet not palatable in its pure form due to its bitter taste. The bitterness diminishes as the hemp juice is mixed with fruit or vegetable juices. The hemp juice simultaneously heightens the fruity, savory and sweet taste of juices and foods in general. Therefore hemp juice is often used in drinks, certain food dishes, seasoning, food supplements, medical products as well as cosmetics. The press remnants can also be used as food supplements.

The approved formulation is given by subcutaneous injection and is used on an as-needed basis rather than on a fixed daily schedule. An intranasal version was studied earlier but did not reach the same stage of development. Route of delivery strongly affects how quickly the peptide appears in circulation.
It means the product is taken shortly before anticipated activity instead of every day. This pattern reduces total exposure and shapes how safety monitoring is planned. It also means effects are expected to be temporary rather than cumulative.
Desire is assessed through validated self-report questionnaires, sometimes paired with event counts and a distress scale. These instruments capture subjective experience, so results depend on how participants interpret the questions. No laboratory marker currently substitutes for such reporting.
Trial results generally show a small to moderate average improvement in desire scores relative to placebo. The distribution of responses is wide, and some participants show little measurable change. Group averages should not be read as a prediction for any single person.
