=== Endoplasmic reticulum === The discovery of an animal cell AMPylase, followed by the discovery of its ER localisation and that BiP is a prominent substrate for its activity were important breakthroughs. BiP (also known as Grp78) had long been known to undergo an inactivating post-translational modification, but its nature remained elusive. Widely assumed to be ADP-ribosylation, it turns out to be FICD-mediated AMPylation, as inactivating the FICD gene in cells abolished all measurable post-translational modification of BiP. BiP is an ER-localised protein chaperone whose activity is tightly regulated at the transcriptional level via a gene-expression program known as the Unfolded Protein Response (UPR). The UPR is a homeostatic process that couples the transcription rate of BiP (and many other proteins) to the burden of unfolded proteins in the ER (so-called ER stress) to help maintain ER proteostasis. AMPylation adds another rapid post-translational layer of control of BiP's activity, as modification of Thr518 of BiP's substrate-binding domain with an AMP locks the chaperone into an inactive conformation. This modification is selectively deployed as ER stress wanes, to inactivate surplus BiP. However, as ER stress rises again, the same enzyme, FICD, catalyses the opposite reaction, BiP de-AMPylation. An understanding of the structural basis of BiP AMPylation and de-AMPylation is gradually emerging, as are clues to the allostery that might regulate the switch in FICD's activity but important details of this process as it occurs in cells remain to be discovered.

=== Promoter escape === RNA polymerase then starts to synthesize the initial DNA-RNA heteroduplex, with ribonucleotides base-paired to the template DNA strand according to Watson-Crick base-pairing interactions. As noted above, RNA polymerase makes contacts with the promoter region. However these stabilizing contacts inhibit the enzyme's ability to access DNA further downstream and thus the synthesis of the full-length product. In order to continue RNA synthesis, RNA polymerase must escape the promoter. It must maintain promoter contacts while unwinding more downstream DNA for synthesis, "scrunching" more downstream DNA into the initiation complex. During the promoter escape transition, RNA polymerase is considered a "stressed intermediate." Thermodynamically the stress accumulates from the DNA-unwinding and DNA-compaction activities. Once the DNA-RNA heteroduplex is long enough (~10 bp), RNA polymerase releases its upstream contacts and effectively achieves the promoter escape transition into the elongation phase. The heteroduplex at the active center stabilizes the elongation complex. However, promoter escape is not the only outcome. RNA polymerase can also relieve the stress by releasing its downstream contacts, arresting transcription. The paused transcribing complex has two options: (1) release the nascent transcript and begin anew at the promoter or (2) re-establish a new 3′-OH on the nascent transcript at the active site via RNA polymerase's catalytic activity and recommence DNA scrunching to achieve promoter escape.
== Fascia research == In 2007, Schleip along with Werner Klingler organized the first Fascia Research Congress, sponsored by the National Institute of Health and hosted at Harvard Medical School. The conference was covered in a two-page Science Magazine article titled "Cell Biology Meets Rolfing with a section titled "From Rolfer to Researcher" referring to Schleip's career shift. All cells in animal body tissues are electrically polarized – in other words, they maintain a voltage difference across the cell's plasma membrane, known as the membrane potential. This electrical polarization results from a complex interplay between protein structures embedded in the membrane called ion pumps and ion channels. In neurons, the types of ion channels in the membrane usually vary across different parts of the cell, giving the dendrites, axon, and cell body different electrical properties. As a result, some parts of the membrane of a neuron may be excitable (capable of generating action potentials), whereas others are not. Recent studies have shown that the most excitable part of a neuron is the part after the axon hillock (the point where the axon leaves the cell body), which is called the axonal initial segment, but the axon and cell body are also excitable in most cases. Each excitable patch of membrane has two important levels of membrane potential: the resting potential, which is the value the membrane potential maintains as long as nothing perturbs the cell, and a higher value called the threshold potential. At the axon hillock of a typical neuron, the resting potential is around −70 millivolts (mV) and the threshold potential is around −55 mV. Synaptic inputs to a neuron cause the membrane to depolarize or hyperpolarize; that is, they cause the membrane potential to rise or fall. Action potentials are triggered when enough depolarization accumulates to bring the membrane potential up to threshold.
