Bremelanotide acts as an agonist at melanocortin receptors, a family of five G-protein-coupled receptors labeled MC1 through MC5. Binding studies indicate activity at several of these subtypes rather than strict selectivity for one. Signalling proceeds mainly through Gs-mediated activation of adenylyl cyclase, raising intracellular cyclic AMP. The MC4 receptor, expressed in hypothalamic and limbic circuits, is widely regarded as the subtype most relevant to sexual response. Because the molecule is not subtype-selective, effects at other melanocortin receptors are expected and are used to explain some observed side effects.
The peptide contains seven amino acids arranged in a ring, closed by a lactam bridge between a side-chain acid and an amine. This cyclic constraint holds the backbone in a defined conformation and increases resistance to enzymatic breakdown relative to linear analogs. N-terminal acetylation and a C-terminal amide further protect the molecule from exopeptidases. The result is a compound with a comparatively long circulation time for a small peptide. Structural modification of the bridge alters receptor affinity, which is one reason analogs in this family differ in their subtype preferences.
Receptor activation in hypothalamic and limbic circuits is the mechanism most often cited for the observed effects on sexual desire. Signalling through MC4R couples to Gs proteins and raises intracellular cyclic AMP, which in turn modulates dopaminergic tone in reward-related pathways. Because the peptide reaches the central nervous system after subcutaneous administration, peripheral vascular changes are regarded as secondary rather than primary. The precise neural circuits that translate receptor occupancy into behavioural change remain incompletely mapped, and published accounts describe the pathway in general terms rather than as a fully resolved sequence.
Development began with intranasal formulations investigated for erectile dysfunction, but blood pressure elevation limited that route and prompted a switch to subcutaneous delivery. Clinical testing then shifted toward hypoactive sexual desire disorder in premenopausal women, and a subcutaneous product received United States approval in 2019. Later trials examined other populations with mixed results, and questions about effect size, durability and patient selection remain open in the peer-reviewed literature. Research interest continues in parallel with the broader melanocortin field, where several synthetic analogues are studied together.

Bremelanotide functions as an agonist at several melanocortin receptor subtypes, with the strongest functional activity reported at the MC4 subtype. MC4 receptors sit in hypothalamic circuits that influence appetite, energy balance, and components of sexual behaviour. Rodents lacking functional MC4 receptors show altered mating behaviour, which supports a role for this pathway in desire. The precise sequence of events connecting receptor activation to reported human effects remains only partly characterised. Because the same receptor family governs pigmentation and inflammatory signalling, selectivity is a recurring theme in pharmacological discussion.
Clinical programmes in this area have relied mainly on randomised, double-blind, placebo-controlled designs in premenopausal women. Primary endpoints usually combine a validated questionnaire covering desire domains with counts of satisfying sexual events and a separate measure of distress. Reported outcomes show statistically significant but modest average improvement over placebo, with wide individual variation. Adverse events such as nausea, flushing, and headache occur frequently and can limit tolerability. Whether short-term trial gains translate into lasting change for most users is an open question.
Evidence outside the studied population is sparse. Trials have concentrated on premenopausal women with a defined diagnosis, and data for postmenopausal women, men, and people taking interacting medications remain limited. Non-prescription use of the peptide for comparable goals is widespread but is not supported by published controlled data. Observed changes in blood pressure have drawn attention to cardiovascular monitoring during use. The literature generally frames the compound as a targeted receptor agonist rather than a general libido enhancer, and basic questions about mechanism and long-term safety are unresolved.
=== Common deprotection cocktails for Fmoc during SPPS === 20% piperidine in DMF (Fmoc group has an approximate half life of 6 seconds in this solution) 5% piperazine, 1% DBU and 1% formic acid in DMF. This method avoids the use of strictly controlled piperidine. No side product was observed for a peptide with 9 residues synthesized with this method.
The sclera, also known as the white of the eye or, in older literature, as the tunica albuginea oculi, is the opaque, fibrous, protective outer layer of the eye containing mainly collagen and some crucial elastic fiber. In the development of the embryo, the sclera is derived from the neural crest. In children, it is thinner and shows some of the underlying pigment, appearing slightly blue. In the elderly, fatty deposits on the sclera can make it appear slightly yellow. People with dark skin can have naturally darkened sclerae, the result of melanin pigmentation. In humans, and some other vertebrates, the whole sclera is white or pale, contrasting with the coloured iris.
Besides OI's association with sensorineural hearing loss, OI is associated with several neurological abnormalities, usually involving the central nervous system, due to deformities in the skeletal structures surrounding it. Neurological complications, especially basilar invagination, may adversely affect life expectancy. In OI, this is most often due to upwards migration of the dens, a feature of the C2 vertebra. Neurosurgery may be needed to correct severe abnormalities when they risk the patient's life or cause either great suffering or intolerable neurological deficits.
Italian publication Everyeye retrospectively described Virtua Tennis as a "killer application" for the Dreamcast, noting the game's considerable commercial success. Gran Turismo 3 and the Grand Theft Auto games are the killer apps for the PlayStation 2. Star Wars Rogue Squadron II: Rogue Leader, Super Smash Bros. Melee, and Super Mario Sunshine are the killer apps for the GameCube. Halo: Combat Evolved, Halo 2, Project Gotham Racing and Dead or Alive 3 are the killer apps for the Xbox. The subsequent Halo series entries became killer apps for the Xbox 360 and Xbox One. Many video game and technology critics call Xbox Live a more general killer app for the Xbox. Dead or Alive 4, the Forza, Gears of War and Mass Effect series are also killer apps for the Xbox 360. Blue Dragon and Dead or Alive 4 were killer apps for the Xbox 360 in Japan. Wii Sports is the killer app for the Wii. Metal Gear Solid 4: Guns of the Patriots boosted PlayStation 3 sales. Mario Kart 8 is a killer app for the Wii U in the UK. The Legend of Zelda: Breath of the Wild is a killer app for the Nintendo Switch. Half-Life: Alyx is a killer app for virtual reality headsets, as the first true AAA virtual reality game. Sales of VR headsets such as the Valve Index increased dramatically after its announcement, suggesting users bought the product specifically for the game. Microsoft Flight Simulator was called a killer app for Xbox Game Studios's Xbox Game Pass subscription, and the Xbox Series X/S.

=== Media === The Houthis are said to have "a huge and well-oiled propaganda machine". They have established "a formidable media arm" with the Lebanese Hezbollah's technical support. The format and content of the group's leader, Abdul-Malik al-Houthi's televised speeches are said to have been modeled after those of Hezbollah's Secretary General, Hassan Nasrallah. Following the peaceful youth uprising in 2011, the group launched its official TV channel, Almasirah. The group operates up to 25 print and electronic newspapers, along with various online news services. One of the most versatile form of Houthi mass media are the zawamil, a genre of primarily tribal oral poetry embedded in Yemen's social fabric. The zamil, rooted in cultural tradition, has been weaponised by the Houthis as a tool of propaganda and remains one of the most popular and rapidly growing platforms of Houthi propaganda, sung by popular vocalists like Issa al-Laith and disseminated through various social media platforms including YouTube, Twitter and Telegram. The Spectator describes Houthi zawamil as its most successful part of their propaganda, stressing the movement's claimed virtues of piety, bravery and poverty in comparison with the corruption, wealth and hypocrisy of their adversaries, the Saudi-led coalition, and Arab states allied to Israel. The Houthis use radio as an effective tool for spreading influence, often seizing stations and confiscating equipment from outlets that fail to comply with their broadcast restrictions.
=== Trials of drugs === Some clinical trials involve healthy subjects with no pre-existing medical conditions. Other clinical trials pertain to people with specific health conditions who are willing to try an experimental treatment. Pilot experiments are conducted to gain insights for design of the clinical trial to follow. There are two goals to testing medical treatments: to learn whether they work well enough, called "efficacy", or "effectiveness"; and to learn whether they are safe enough, called "safety". Neither is an absolute criterion; both safety and efficacy are evaluated relative to how the treatment is intended to be used, what other treatments are available, and the severity of the disease or condition. The benefits must outweigh the risks. For example, many drugs to treat cancer have severe side effects that would not be acceptable for an over-the-counter pain medication, yet the cancer drugs have been approved since they are used under a physician's care and are used for a life-threatening condition. In the US the elderly constitute 14% of the population, while they consume over one-third of drugs. People over 55 (or a similar cutoff age) are often excluded from trials because their greater health issues and drug use complicate data interpretation, and because they have different physiological capacity than younger people. Children and people with unrelated medical conditions are also frequently excluded. Pregnant women are often excluded due to potential risks to the fetus.
Eduard Buchner submitted his first paper on the study of yeast extracts in 1897. In a series of experiments at the University of Berlin, he found that sugar was fermented by yeast extracts even when there were no living yeast cells in the mixture. He named the enzyme that brought about the fermentation of sucrose "zymase". In 1907, he received the Nobel Prize in Chemistry for "his discovery of cell-free fermentation". Following Buchner's example, enzymes are usually named according to the reaction they carry out: the suffix -ase is combined with the name of the substrate (e.g., lactase is the enzyme that cleaves lactose) or to the type of reaction (e.g., DNA polymerase forms DNA polymers). The biochemical identity of enzymes was still unknown in the early 1900s. Many scientists observed that enzymatic activity was associated with proteins, but others (such as Nobel laureate Richard Willstätter) argued that proteins were merely carriers for the true enzymes and that proteins per se were incapable of catalysis. In 1926, James B. Sumner showed that the enzyme urease was a pure protein and crystallized it; he did likewise for the enzyme catalase in 1937. The conclusion that pure proteins can be enzymes was definitively demonstrated by John Howard Northrop and Wendell Meredith Stanley, who worked on the digestive enzymes pepsin (1930), trypsin and chymotrypsin. These three scientists were awarded the 1946 Nobel Prize in Chemistry. The discovery that enzymes could be crystallized eventually allowed their structures to be solved by x-ray crystallography.

== Influences and style == He described himself as a very inquisitive child saying "I enjoyed catching and keeping living things, such as fish or birds. Other than that, I liked both watching and making animation. Basically, I seemed to be interested in things that moved." Among his favorite subjects in school was art. He commented, "If I was not in the games industry, I would want to become a classical artist. Though I regard not only games but also anything that expresses something – be it films, novels or manga – as forms of art." Ueda played many Sega Mega Drive games, which influenced his work. He was also a fan of the Amiga computer platform games Flashback and Another World during his teen years. Other games that influenced his work include The Legend of Zelda, Virtua Fighter, and Prince of Persia. He was also influenced by the work of Kenji Eno, and the manga series Galaxy Express 999 (1977–1981). Ueda's games are considered to have a distinctive style, which Ueda himself describes as "design by subtraction", with sparse landscapes, oversaturated lighting and minimalist story to give his games a personal and distinctive feel. Ueda also said that, in video games, ideas for a gameplay mechanic should be made first, then complemented by a game's story.
==== Indigenous Americans ==== Aztec paintings, Central American temples, and the great burial mounds in the Southern United States are frequently adorned with depictions of rattlesnakes, often within the symbols and emblems of the most powerful deities. The Feathered Serpent of Mesoamerican religion was depicted as having the combined features of the quetzal and rattlesnake. The Ancient Maya considered the rattlesnake to be a "vision serpent" that acted as a conduit to the "otherworld". Rattlesnakes are a key element in Aztec mythology and were widely represented in Aztec art, including sculptures, jewelry, and architectural elements.
LMNA missense mutation (c.1580G>T) introduced at LMNA gene – position 1580 (nt) in the DNA sequence (CGT) causing the guanine to be replaced with the thymine, yielding CTT in the DNA sequence. This results at the protein level in the replacement of the arginine by the leucine at the position 527. This leads to destruction of salt bridge and structure destabilization. At phenotype level this manifests with overlapping mandibuloacral dysplasia and progeria syndrome. The resulting transcript and protein product is:

It is frequently described as an MC4 receptor agonist, but binding assays show activity at more than one melanocortin subtype. Selectivity is therefore relative rather than absolute. This matters when interpreting side effects tied to other receptor subtypes.
The lactam bridge locks the peptide into a defined shape and makes it harder for peptidases to cut the backbone. Terminal blocking groups add further protection at both ends. Together these features extend the molecule's persistence in circulation compared with linear peptides.
Evidence from animals favors hypothalamic melanocortin circuits, yet direct confirmation in humans is lacking. A peripheral vascular contribution has also been proposed. How receptor binding translates into a reported behavioral effect is still not mapped.
The code designates bremelanotide, a cyclic heptapeptide melanocortin receptor agonist. It served as an internal research identifier before the compound entered formal clinical development. The code and the generic name refer to the same molecule.
