However, the X-ray diffraction structure may not exist, and, since the proteins in solution are flexible molecules, a protein represented by a single structure may lead to underestimate the intrinsic variation of the atomic positions of a protein. A set of conformations, determined by NMR or X-ray crystallography may be a better representation of the experimental data of a protein than a unique conformation. The utility of a model will be given, at least in part, by the degree of accuracy and precision of the model. An accurate model with relatively poor precision could be useful to study the evolutionary relationships between the structures of a set of proteins, whereas the rational drug design requires both precise and accurate models. A model that is not accurate, regardless of the degree of precision with which it was obtained will not be very useful. Since protein structures are experimental models that can contain errors, it is very important to be able to detect these errors. The process aimed at the detection of errors is known as validation. === Key challenges === LEDs require optimized efficiency to hinge on ongoing improvements such as phosphor materials and quantum dots. The process of down-conversion (the method by which materials convert more-energetic photons to different, less energetic colors) also needs improvement. For example, the red phosphors that are used today are thermally sensitive and need to be improved in that aspect so that they do not color shift and experience efficiency drop-off with temperature. Red phosphors could also benefit from a narrower spectral width to emit more lumens and becoming more efficient at converting photons. In addition, work remains to be done in the realms of current efficiency droop, color shift, system reliability, light distribution, dimming, thermal management, and power supply performance. Early suspicions were that the LED droop was caused by elevated temperatures. Scientists showed that temperature was not the root cause of efficiency droop. The mechanism causing efficiency droop was identified in 2007 as Auger recombination, which was taken with mixed reaction. A 2013 study conclusively identified Auger recombination as the cause.

== Pharmacology == Coronaridine has been reported to bind to an assortment of molecular sites, including: μ-opioid (Ki = 2.0 μM), δ-opioid (Ki = 8.1 μM), and κ-opioid receptors (Ki = 4.3 μM), NMDA receptor (Ki = 6.24 μM) (as an antagonist), and nAChRs (as an antagonist). It has also been found to inhibit the enzyme acetylcholinesterase, act as a voltage-gated sodium channel blocker, and displays estrogenic activity in rodents. In contrast to ibogaine and other iboga alkaloids, coronaridine does not bind to either the σ1 or σ2 receptor.
== Public health concerns == Epidemics of fatal pulmonary hypertension and heart valve damage associated with pharmaceutical anorectic agents have led to the withdrawal of products from the market. This was the case with aminorex in the 1960s, and again in the 1990s with fenfluramine (see: Fen-phen). Likewise, association of the related appetite suppressant phenylpropanolamine with hemorrhagic stroke led the Food and Drug Administration (FDA) to request its withdrawal from the market in the United States in 2000, and similar concerns regarding ephedrine resulted in an FDA ban on its inclusion in dietary supplements in 2004. A Federal judge later overturned this ban in 2005 during a challenge by supplement maker Nutraceuticals. It is also debatable as to whether the ephedrine ban had more to do with its use as a precursor in methamphetamine manufacture rather than health concerns with the ingredient as such.
=== Neonicotinoids === Neonicotinoids are a class of neuro-active insecticides chemically similar to nicotine, with much lower acute mammalian toxicity and greater field persistence. These chemicals are acetylcholine receptor agonists. They are broad-spectrum systemic insecticides, with rapid action (measured in minutes or hours). They are applied as sprays, drenches, seed and soil treatments. Treated insects exhibit leg tremors, rapid wing motion, stylet withdrawal (aphids), disoriented movement, paralysis and death. Imidacloprid, of the neonicotinoid family, is the most widely used insecticide in the world. In the late 1990s neonicotinoids came under increasing scrutiny over their environmental impact and were linked in a range of studies to adverse ecological effects, including honey-bee colony collapse disorder (CCD) and loss of birds due to a reduction in insect populations. In 2013, the European Union and a few non EU countries restricted the use of certain neonicotinoids. and its potential to increase the susceptibility of rice to planthopper attacks.
