Bremelanotide is a moderately large peptide with a molecular mass near 1025 daltons. In lyophilised form it appears as a white to off-white powder and is freely soluble in water and other polar solvents. The intact lactam ring is essential for receptor affinity, while linearised fragments bind far more weakly. Solutions are sensitive to extremes of pH and to prolonged exposure to light and heat, so handling typically involves buffered conditions and cold storage. Its short plasma half-life reflects rapid distribution and clearance rather than chemical breakdown inside the vial.
Development of the peptide passed through several delivery formats, including an intranasal version tested in early trials and an injectable version that entered later clinical study. Regulatory approval for a subcutaneous product in the United States was granted in 2019 after review of controlled trials in premenopausal women. == Treatment == No cure for scleroderma is known, though treatments exist for some of the symptoms, including drugs that soften the skin and reduce inflammation. Some patients may benefit from exposure to heat. Holistic care of patients comprising patient education tailored to patients' education level is useful in view of the complex nature of the disease symptoms and progress.

== History == Insulin was discovered by Sir Frederick G Banting, Charles H Best, and JJR Macleod from the University of Toronto in 1921 as an injectable agent. German researchers first introduced the idea of inhalable insulin in 1924. Years of failure followed until scientists realized they might be able to use new technologies to turn insulin into a concentrated powder with particles sized for inhalation. In the 1980s Nektar Therapeutics, based on work by A. Carl Leopold on vitrifying proteins, developed technology to make insulin into small particles, technology then licensed to Pfizer. Alkermes developed a delivery device that they licensed to Eli Lilly and Company. Once concrete methods were developed, human tests began in the late 1990s. In January 2006, the U.S. Food and Drug Administration (FDA) approved the use of Exubera, a form of inhalable insulin developed by Pfizer. It was approved in the UK in August 2006 but reimbursed by the National Health Service only for people who had problems with needles. It was not reimbursed by any U.S. insurer. A 2007 systematic review concluded that the inhaled hexameric insulin (Exubera) "appears to be as effective, but no better than injected short-acting insulin. The additional cost is so much more that it is unlikely to be cost-effective." In 2007, Pfizer announced that it would no longer manufacture or market Exubera. According to Chairman and CEO Jeffrey Kindler this was because Exubera "failed to gain acceptance among patients and physicians".

Immature granulocyte Immature granulocytes are immature forms of neutrophils and other granulocytes (eosinophils and basophils). This classification consists of metamyelocytes, myelocytes and promyelocytes, which may be enumerated separately in the manual differential or reported together as immature granulocytes (IG) by automated methods. Immature granulocytes are normally found in the bone marrow, but not in peripheral blood. When present in significant quantities in the blood, immature granulocytes can indicate infection and inflammation, as well as myeloproliferative disease, leukaemia and other conditions affecting the marrow. IGs may also be increased in steroid use and pregnancy. Chronic myeloid leukaemia often presents with a high number of immature granulocytes in the peripheral blood. Abnormal promyelocytes with multiple Auer rods, called faggot cells, occur in acute promyelocytic leukaemia.

A few weeks prior to Operation Protea, SADF General Charles Lloyd warned Botha that the introduction of early-warning radar and 2K12 Kub "SA-6" missiles in southern Angola was making it difficult to provide air support to ground operations there. Lloyd mentioned that FAPLA's buildup of modern Soviet arms was making a conventional war more likely. The objectives of Operation Protea shifted accordingly: aside from the PLAN camps, the SADF was ordered to neutralise several Angolan radar and missile sites and command posts. Eight days of bloody fighting occurred before two South African armoured columns were able to overrun Ondjiva and Xangongo. The SADF destroyed all of FAPLA's 2K12 missile sites and captured an estimated 3,000 tonnes of Soviet-manufactured equipment, including a dozen T-34-85 and PT-76 tanks, 200 trucks and other wheeled vehicles, and 110 9K32 Strela-2 missile launchers. The SADF acknowledged 14 dead. Combined FAPLA and PLAN losses were over 1,000 dead and 38 taken prisoner. The Soviet military mission suffered 2 dead and 1 taken prisoner. Operation Protea led to the effective occupation of forty thousand square kilometres of Cunene Province by the SADF. On 31 August, the US vetoed a UN Security Council resolution condemning the incursion and demanding the immediate and unconditional withdrawal of the SADF from Angola. Intelligence gained during Protea led to Operation Daisy in November 1981, the deepest SADF incursion into Angola since Operation Savannah.
