== Mechanism == The enzyme uses pyridoxal phosphate (PLP), the active form of vitamin B6, as a cofactor. PLP is essential to the mechanism of decarboxylation in AADC. In the active enzyme, PLP is bound to lysine-303 of AADC as a Schiff base. Upon substrate binding, Lys-303 is displaced by the substrate's amine. This positions the carboxylate of the substrate within the active site such that decarboxylation is favored. Decarboxylation of the substrate produces a quinonoid intermediate, which is subsequently protonated to produce a Schiff base adduct of PLP and the decarboxylated product. Lys-303 can then regenerate the original Schiff base, releasing the product while retaining PLP. Probing this PLP-catalyzed decarboxylation, it has been discovered that there is a difference in concentration and pH dependence between substrates. DOPA is optimally decarboxylated at pH 6.7 and a PLP concentration of 0.125 mM, while the conditions for optimal 5-HTP decarboxylation were found to be pH 8.3 and 0.3 mM PLP.

Many psychoactive drugs can be or have been specifically designed for sublingual administration, including barbiturates, benzodiazepines, opioid analgesics with poor gastrointestinal bioavailability, LSD blotters, coca leaves, some hallucinogens. The 62-ton Ivy Mike device built by the United States and exploded on 1 November 1952, was the first fully successful hydrogen bomb (thermonuclear bomb). In this context, it was the first bomb in which most of the energy released came from nuclear reaction stages that followed the primary nuclear fission stage of the atomic bomb. The Ivy Mike bomb was a factory-like building, rather than a deliverable weapon. At its center, a very large cylindrical, insulated vacuum flask or cryostat, held cryogenic liquid deuterium in a volume of about 1000 liters (160 kilograms in mass, if this volume had been completely filled). Then, a conventional atomic bomb (the "primary") at one end of the bomb was used to create the conditions of extreme temperature and pressure that were needed to set off the thermonuclear reaction. Within a few years, so-called "dry" hydrogen bombs were developed that did not need cryogenic hydrogen. Released information suggests that all thermonuclear weapons built since then contain chemical compounds of deuterium and lithium in their secondary stages. The material that contains the deuterium is mostly lithium deuteride, with the lithium consisting of the isotope lithium-6. When the lithium-6 is bombarded with fast neutrons from the atomic bomb, tritium (hydrogen-3) is produced, and then the deuterium and the tritium quickly engage in thermonuclear fusion, releasing abundant energy, helium-4, and even more free neutrons. "Pure" fusion weapons such as the Tsar Bomba are believed to be obsolete.

IGF-1 levels are significantly lower in cats with untreated diabetes mellitus without hypersomatotropism. To counteract this, diabetic cats with suspected hypersomatotropism can be treated for 6–8 weeks with insulin before testing. Another issue with IGF-1 testing is that most tests consider values of other 1000 ng/mL to be indicative of hypersomatotropism, even though healthy cats and cats with diabetes mellitus but not hypersomatotropism have levels below 800 ng/mL, leaving a grey zone of 800–1000 ng/mL. A study in 2000 reported eight cats with diabetes mellitus without hypersomatotropism had levels of IGF-1 above the normal range. Other studies suggested that this result was due to long term insulin therapy. Technical issues with the testing may result in false reports of increased levels due to the tests removing the proteins that circulating IGF-1 binds to. Multiple cats with IGF-1 levels reported above 1000 ng/mL did not show signs of hypersomatotropism during further examination.
== Origin == There are conflicting stories about the dessert's origin. Some claim that it was first served during the 1893 Chicago World's Fair, where the ice cream sundae was also invented. Though in 1894, a Philadelphia company was given credit for its invention describing: "A small, solid [cake] of the ice cream is enveloped in a thin sheet of pie crust and then dipped into boiling lard or butter to cook the outside to a crisp. Served immediately, the ice cream is found to be as solidly frozen as it was first prepared." It may be derived in part from Baked Alaska, originating earlier in the 19th century, which puts ice cream under a quickly broiled meringue.
== Biological Role of cGP == The hepatic production of IGF-1 is controlled by the growth hormone (GH)-IGF-1 axis. The majority of circulating IGF-1 is not bioavailable because of its affinity and binding to IGF-binding protein (IGFBP), mainly IGFBP3. IGF-1 bioactivity is therefore, tightly regulated through reversible binding with IGFBP3. It is this binding-release process that determines the amount of bioavailable IGF-1 in circulation. IGF-1 that is not bound, is cleaved into an N-terminal tripeptide, glycine-proline-glutamate (GPE) and Des-N-IGF-1. and GPE metabolizes to result in cyclic glycine proline (cGP). Unbound IGF-1, cleaved at the N-terminal, can be metabolized through a series of downstream enzymatic reactions to cGP. The N-terminal is the binding site of IGF-1 which allows cGP to retain the same binding affinity to IGFBP-3 and thus, regulates the bioavailability of IGF-1 through competitive binding with IGFBP3. An increase in cGP, would increase competitive advantage and thus, increase the amount of circulating and therefore, bioavailable IGF-1. Research shows that cGP can normalize IGF-1 function under pathophysiological conditions of increased or diminished IGF-1 bioactivity. In vitro studies show that cGP promoted the activity of IGF-1 when insufficient and inhibited the activity of IGF-1 when in excess.
